چکیده

Eggs of the sea urchin Arbacia punctulata were collected by the electroshock method (1), washed in artificial seawater, and sedimented by centrifugation at 500 g for 3 min . Embryos were obtained by fertilizing eggs in artificial seawater with a drop of fresh dilute sperm suspension . Only those cultures in which fertilization was 95-1007 effective were used for the isolation of nuclei. The sedimented cells were resuspended in 9 volumes of a 1 M sucrose solution containing 2% (w/v) of the nonionic detergent Triton X-100 and 2 MM MgCI2. Subsequent steps were monitored by light microscopy. The suspension was homogenized in a Potter-Elvehjem homogenizer with 15 slow upand-down strokes of a loose Teflon pestle . The homogenate contained cytoplasmic debris, occasional unbroken cells, and fragments of fertilization membrane when unhatched embryos were used . The homogenate was next centrifuged at 2000 g for 30 min, the supernatant discarded, and the detergent wash repeated . The sediment, containing nuclei and membranes, was then suspended in 3 volumes of 1 .4 mI sucrose, and layered on a discontinuous gradient composed of 1 ml of 2.5 mI sucrose layered on the bottom, 1 ml of 2 .2 mI sucrose in the middle layer, and 1 ml of 1 .9 M sucrose on the top . The preparation was then centrifuged at 25,000 rpm for 45 min at 4°C in a Spinco SW-39 swinging bucket rotor . The layers containing lysed cytoplasm were carefully removed from the top, with a pasteur pipette .

The Isolation of Nuclei from Eggs and Embryos of the Sea Urchin

NUCLEI CAN BE ISOLATED FROM THE EGGS AND EMBRYOS OF THE SEA URCHIN BY A FOUR STEP METHOD: (1) washing with dextrose solution until the cells become weak, (2) cytolysis in 0.002 M Mg(++) solution, (3) homogenization of the cytolyzed cells by agitation, and (4) separation of the nuclei from the rest of the homogenate by centrifugation on a layered sucrose gradient. Contamination of the isolated n...

Determination of developmental stages of embryo in the Sea Urchin, Echinometra mathaei

Sea Urchin is one of the most useful tools in developmental biology studies because this organism has the simplest kind of developmental stages. We aimed to determine developmental stages and timetable of Echinometra mathaei embryo (the species of Persian Gulf). The spawning of E. mathaei was induced by 0.5M KCl injection (1ml) into the coelomic cavity. After fertilization, embryos were placed ...

Embryonic, larval and juvenile development of tropical sea urchin, Diadema setosum

Diadema setosum (Leske, 1778), is one of the common echinoids widely distributed in the Indo-West Pacific Ocean, where it occurs from the Red Sea, Persian Gulf and the east coast of Africa to Japan, Australia and Malaysia. To investigate the developmental basis of morphological changes in embryos and larvae, we documented the ontogeny of D. setosum in a controlled laboratory condition at the In...

Post-transcriptional restrict!on of gene expression m sea urchin interspecies hybrid embryos

The synthesis of many paternal species-specific proteins is reduced in all stages of sea urchin interspecies hybrid embryos, due to the reduced amounts of some paternal mRNA species in hybrid embryos compared with the embryos of the paternal species (Tufaro and Brandhorst 1982). Possible explanations for this restriction were tested. Cloned cDNAs were selected that were specific for paternal RN...
